The Cold Blow Lane Upper and Barry Kitchener Stand only will be open for the game. Tickets for Juniors are just £1 and are available right up until kick-off. Adults are £10 and tickets for Over 60s are £5.
Should the score be level at the end of 90 minutes, the tie will go straight to penalties.
Carlos Edwards could be in line for his first competitive appearance in just under 12 months after signing a new short-term deal with Millwall last week.
The experienced right-back has successfully overcome a serious knee injury picked up in September 2014 and, having proved his fitness to boss Neil Harris since the start of pre-season, could be in contention for what will be an emotional first team return at The Den.
The game may come too for Joe Martin, who has only just returned to training following an ankle injury, while Shane Ferguson (hamstring) remains sidelined.
Posh are without central defenders Gabi Zakuani (suspension) and Jack Baldwin (injured) and are also waiting on the fitness of full-backs Kgosi Ntlhe and Michael Smith.
New recruit Chris Forrester is in line to feature while Callum Elder will continue should Ntlhe be unable to play.
Tuesday night's meeting will be the 29th between Millwall and Peterborough United.
The Lions have enjoyed a better head-to-head record so far, picking up 13 wins to Posh's nine, while six games have ended in draws.
Four of the previous 28 matches have come knockout competition - specifically the League Cup - where both Millwall and Peterborough have two victories each.
